百合GARP家族鉴定和鳞片发育的基因克隆与表达

GARP是植物特异性转录因子家族,在植物生长发育和非生物胁迫响应中发挥关键作用.为全面了解百合GARP转录因子提供基础资料,为探索百合鳞片的生长发育机制奠定一定基础,基于二代、三代百合转录组数据,对其GARP家族成员进行鉴定和表达分析.结果表明,在转录组数据中共鉴定出34个GARP蛋白,均为亲水性蛋白,68%为酸性蛋白,94%为不稳定蛋白.系统进化树将百合GARPs分为 5个亚家族:ARR-B、GLK、NIGT1/HRS1/HHO、PHR1/PHL1和KAN,进化分析发现,百合GARPs可能参与生长发育、生物钟调节、开花转化、激素运输、非生物胁迫等过程.对鳞片发育具有重要作用的ARR-B亚家族成员进行qRT-PCR分析,结果发现,LhGARP33、LhGARP30和LhGARP3可能对鳞茎发育起重要作用.LhGARP3和LhGARP30被成功克隆,氨基酸序列具有典型的ARR-Bs磷酸受体结构域(REC)及金属和磷酸化结合位点.空间表达分析了ARR-B I亚组在花、叶和鳞片中的表达模式,结果表明,LhGARP5在叶中的表达量最高,而LhGARP31在花中的表达量最高,LhGARP33、LhGARP30和LhGARP3的表达特征基本一致,均在鳞片中高表达,其可能作为关键基因在鳞片发育过程中发挥主要作用.
Identification of GARP Transcription Factor Family and Cloning and Expression of Bulb Development Related Genes in Lilium
GARP is a plant specific transcription factor family and plays a key role in plant growth,development,and abiotic stress response.In order to provide basic information for a comprehensive understanding of GARP transcription factors in Lilium,and laid a foundation for exploring the growth and development mechanism of bulbs in Lilium,in this study,based on the transcriptome data of Next-generation sequencing and Third-generation sequencing of Lilium,identification and expression analysis of GARP family members were conducted.The results indicated that a total of 34 GARP proteins were identified in transcriptome data,all the GARP proteins were hydrophilic,68%of which were acidic proteins,and 94%were unstable proteins.Based on the phylogenetic analysis,Lilium GARPs were clustered into five subfamilies:ARR-B,GLK,NIGT1/HRS1/HHO,PHR1/PHL1,and KAN,and they were annotated with functions on growth and development,biological clock regulation,flowering transformation,hormone transport,abiotic stress and other processes.The ARR-B subfamily members that played important roles in bulb development were analyzed by qRT-PCR,and the results showed that LhGARP33,LhGARP30,and LhGARP3 may play important roles in bulb development.The LhGARP3 and LhGARP30 were successfully cloned and their amino acid sequences exhibited the typical ARR-Bs phosphate receptor domain(REC)and metal and phosphorylation binding sites.Spatial expression patterns of ARR-B I subgroup in flowers,leaves,and bulbs were analyzed and the results revealed that LhGARP5 had the highest expression in leaves,while LhGARP31 had the highest expression in flowers,LhGARP33,LhGARP30 and LhGARP3 had the basically consistent expression characteristics,the highest expression in bulbs,indicating that they may play a major role in bulb development as a key gene.
