Objective To describe the composition and structure of esophageal microbiota of tumor and non-tumor tissues in patients with esophageal squamous cell carcinoma(ESCC)and to screen the differences between the two groups.Methods The cancer tissues and adjacent tissues of 48 cases of primary ESCC diagnosed by pathology in Anyang Cancer Hospital were collected,and 5R 16S rRNA getne(V2,V3,V5,V6,V8)sequencing was performed.Wilcoxon rank sum test was used to compare Alpha diversity and relative abundance between the two groups.Results The Alpha diversity of ESCC cancer tissue was higher than that of non-tumor control group.Fir-micutes,Bacteroidota,Proteobacteria and Fusobacteriota were the main phyla in ESCC and non-tumor control groups.At the phylum level,the ESCC group contained higher abundances of Fuso-bacteriota and Spirochaetes,and lower abundances of Actinobacteria and Cyanobacteria(P<0.05)compared with the non-tumor control group.At the genus level,the abundance of Fusobacterium,Capnocytophaga,Ca-tonella and Treponema in ESCC group were significantly higher than that in non-tumor control group,while the abundance of Staphylococcus and Pseudomonas in ESCC group were lower than that in non-tumor control group(P<0.05).At the species level,the abundance of Fusobacterium nucleatum,Fusobacterium periodontic-um,Capnocytophaga sputigena and Porphyromonas gingivalis in ESCC group were higher,while the abundance of Streptococcus dysgalactiae and Sphingomonas leidyi were lower than that in non-tumor control group.Conclusion There were significant differences in esophageal microbiota composition between tumor and adjacent normal tumor tissue samples from ESCC patients.
