Objective To construct CAR-T cells targeting B7H3 and study its inhibitory effect on KYSE150 cells of esophageal carcinoma.Methods The expression of B7H3 in esophageal cancer tissues and adjacent tissues was detected by immunohistochemistry,and the correlation between the expression of B7H3 protein in esophageal cancer tissues and clinicopathological features of patients was analyzed.Lentiviral vector was used to prepare B7H3-CAR-T cells,and the stability efficiency of CAR-T cells was observed by fluorescence mi-croscope.The killing ability of CAR-T cells to KYSE150 cells was detected by co-culture with different efficiency target ratio.The cytokines released in the supernatant were detected by ELISA kit.Results The expression of B7H3 in esophageal carcinoma tissues was significantly higher than that in adjacent tissues,and the expres-sion of B7H3 protein was significantly correlated with the depth of invasion,pTNM stage,lymph node metas-tasis and length of lesion(P<0.05).There was no correlation with gender,age,smoking,drinking,location of tumor and differentiation degree(P>0.05).The novel CAR-T cells with optimized structure were obtained by lentivirus infection.When the effect-target ratio was 10∶1,B7H3-CAR-T cells showed stronger killing a-bility,and the secretion levels of IL-2 and IFN-γ were significantly higher than that of T cells at different effect-target ratios.Conclusion B7H3-targeted CAR-T cells were successfully prepared,which can effec-tively kill B7H3-positive esophageal cancer KYSE150 cells.
