蓝莓开放式无糖组培技术体系建立

以蓝莓无菌试管苗为试材,采用L9(34)正交等试验方法,开展抑菌剂种类和浓度、培养液浓度与基质的比例、CO2浓度等无糖培养主要技术环节研究。结果表明:(1)在基质培养液中添加清菌易8 mL/L,基质中培养液含量80%时,可明显抑制培养基质污染,污染率可控制在15%以内,且蓝莓苗生长、增殖正常。(2)正交试验结果方差分析和极差分析表明,培养液中基本培养基(WPM)比例(1/2 WPM、1/4 WPM、1/8 WPM)对增殖率、污染率、生根率、成苗率、根长影响差异达到极显著水平,CO2浓度对增殖率、生根率、根长影响差异达到极显著水平,激素配比对蓝莓开放式培养的增殖率、生根率等影响也达到差异极显著水平。基质培养液浓度及比例对污染率、生根率等的影响达到差异显著水平。结合考虑降低污染,蓝莓无糖培养的最佳增殖培养配方是培养液为1/4 WPM,CO2浓度1 000-1 500 μmol/mol,珍珠岩基质中培养液含量80%,激素配比(mg/L)ZT:IBA为(0。5-1):2。(3)培养液添加方式试验表明,培养液先低后高的添加方式可降低污染率,明显提高增殖率。
Establishment of open sugar free tissue culture technology system of blueberry
The main technical aspects of sugar-free culture,such as the type and concentration of bacteriostatic agent,the concentration of liquid medium and the ratio of it to the matrix,and CO2 concentration,were carried out by using blueberry sterile seedlings as materials and L9(34)orthogonal experimental methods.The results showed that(1)When 8 mL/L of bacteriostatic agent"Qingjunyi"was added to the matrix culture medium,and the content of the culture medium in the substrate was 80%,the contamination rate could be controlled within 15%,the growth and proliferation of blueberry seedlings were normal.(2)Orthogonal experimental results ANOVA and range analysis showed that the difference of basic medium(WPM)ratio(1/2 WPM,1/4 WPM,1/8 WPM)in the culture medium on proliferation rate,pollution rate,rooting rate,seedling rate and root length reached a extremely significant level(P<0.01,the same below),CO2 concentration on proliferation rate,rooting rate and root length difference reached an extremely significant level,and the effect of hormone ratio on the proliferation rate and rooting rate of blueberry open culture also reached an extremely significant level.The effects of matrix culture concentration and proportion on contamination rate and rooting rate reached a significant level(P<0.05).Combined with the consideration of reducing pollution,the best proliferation culture formula for sugar-free culture of blueberries was 1/4 WPM,CO2 concentration of 1 000-1 500 μmom/mol,culture medium content of 80%in perlite matrix,hormone ratio of ZT:IBA was(0.5-1):2.(3)The test of the addition method of culture medium showed that the addition method of low and then high culture medium could reduce the contamination rate and significantly improve the proliferation rate.
