Research of toxic production conditions for Alternaria ochroleuca using response surface methodology
The toxin production ability of different Alternaria(Alternaria ochroleuca,Alternaria alternata and Alternaria tenuissima)was compared.The effects of culture temperature,culture time,and initial pH of the culture medium on the production of 3 major toxins,namely,alternariol(AOH),alternariol monomethyl ether(AME)and tenuazonic acid(TeA)of A.ochroleuca,were investigated through single factor experiments and response surface methodology.The results showed that the production of 3 toxins showed a trend of first increasing and then gradually decreasing with the increase of culture temperature,culture time,and initial pH of the culture medium.The optimal production conditions for TeA were culture temperature 26.8 ℃,culture time 11.4 days,and initial pH of the culture medium 4.7,with a maximum production of 4 841.3 mg/kg.When the culture temperature was 25.9 ℃,the culture time was 7.8 days,and the initial pH of the culture medium was 6.4,the production of AOH and AME was the highest,reaching 2 096.1 mg/kg and 152.8 mg/kg,respectively.The culture temperature and initial pH of the culture medium had a significant interaction effect on the production of TeA,while the culture temperature and culture time had a significant interaction effect on the production of AOH and AME.This study identified the main influencing factors of A.ochroleuca producing Alternaria toxin,which was of great significance for controlling Alternaria toxin contamination and ensuring food safety.
