Effects of procyanidin addition on rooster semen freezing
In order to explore the application effects of antioxidant procyanidin(PC)in rooster semen freezing,different concentrations of PC(10 µg/mL,20 μg/mL,30 µg/mL,40 µg/mL,60 µg/mL)were added to 10%yolk supernatant dilution,the quality indexes and antioxidant indexes of frozen sperm were detected.The results showed that compared to the control,there was no significant difference in the quality indexes(survival rate,viability,membrane integrity,acrosome integrity,and mitochondrial activity)of frozen rooster sperm when 10-30 μg/mL PC was added to the yolk supernatant dilution.When adding PC with 40-60 μg/mL,all quality indexes of frozen chicken sperm significantly decreased(P<0.05).In terms of antioxidant indexes,the addition of PC could improve the antioxidant capacity of frozen rooster sperm,among which the antioxidant capacity of frozen chicken sperm with 30 μg/mL PC addition was the highest.The MDA content and ROS content of frozen rooster sperm were the lowest,at 5.48 nmol/L and 342.00 U/mL,respectively;SOD activity and GSH-PX activity were the highest,at 137 U/mL and 90.85 U/L,respectively,with significant differences compared to other groups.When the concentration of PC added was 40-60 µg/mL,the antioxidant capacity of frozen rooster sperm significantly decreased,showing toxic side effects on sperm.It could be seen that adding 10-30 μg/mL PC during the freezing process of rooster semen may not significantly improve the quality of frozen rooster sperm,but it can significantly enhance its antioxidant capacity.
