甘薯IbTOE3-like基因的克隆与表达分析

以甘薯品种'西瓜红'为材料,通过同源克隆得到IbTOE3-like基因的全长CDS序列,分析其编码蛋白的理化性质和基本功能。结果表明:IbTOE3-like基因在甘薯块根中的转录水平相较于其他组织中更高,并且伴随甘薯块根的发育而升高,确定其为甘薯块根膨大相关基因。IbTOE3-like蛋白拥有两个相连的AP2结构域,属于AP2/ERF家族的一员,预测其为亲水性蛋白。烟草瞬时转化试验确定IbTOE3-like蛋白定位在细胞核。转录激活活性验证发现,IbTOE3-like基因无转录激活活性。此外,在烟草叶片中瞬时过量表达miR172能有效抑制IbTOE3-like基因的转录水平,并通过5'RACE试验证明了 miR172与IbTOE3-like基因的靶向关系。试验结果可为进一步研究IbTOE3-like基因在甘薯根系膨大过程中的作用奠定基础。
Cloning and expression analysis of IbTOE3-like gene in sweet potato
Using the sweet potato variety'Xiguahong'as the material,the full-length CDS sequence of the IbTOE3-like gene was obtained through homologous cloning,and the physicochemical properties and basic functions of its encoded protein were analyzed.The results showed that the transcription level of IbTOE3-like gene was higher in sweet potato root tuber compared to other tissues,and increased with the development of sweet potato root tuber confirming it as a sweet potato root tuber swelling related gene.The IbTOE3-like protein had two connected AP2 domains and belonged to the AP2/ERF family.It was predicted to be a hydrophilic protein.The tobacco transient transformation test determined that the IbTOE3-like protein was located in the nucleus.Transcriptional activation activity validation revealed that the IbTOE-like gene had no transcriptional activation activity.In addition,transient overexpression of miR172 in tobacco leaves could effectively inhibit the transcription level of IbTOE3-like gene,and the targeted relationship between miR172 and IbTOE3-like gene was demonstrated through the 5'RACE assay.The results could lay the foundation for further research on the role of IbTOE3-like gene in the process of sweet potato root tuber swelling.
