摘要

随着全球人口的老年化,包括骨质疏松症(osteoporosis,OP)在内的年龄相关性疾病发病率逐渐上升,由OP导致的脆性骨折给社会和家庭带来了沉重的负担.应用抗OP药物可以降低骨质疏松性骨折(osteoportic fracture,OPF)的发生,但目前上市的药物存在应用疗程受限及各种副作用,因此寻找更好的防治OP药物势在必行.随着年龄的增长,衰老细胞(senescent cells,SC)在骨微环境中累积,清除这些衰老细胞或者减少衰老相关分泌表型(senescence-associated secretory phenotype,SASP)可预防或减缓OP.这一疗法或可开启OP治疗新模式,并有益于多种与细胞衰老相关的老年性疾病.本文就近年来抗细胞衰老疗法在细胞、动物、人体层面上防治OP取得的研究进展进行综述.

Abstract

With the aging of the global population,the incidence of age-related diseases including osteoporosis is increasing.Fragility fractures caused by osteoporosis have brought a heavy burden to society and families.Using anti-osteoporosis drugs to reduce the occurrence of osteoportic fracture is an ideal approach,but the currently used drugs have various deficiencies,so it is imperative to find better anti-osteoporosis drugs for the prevention and treatment of osteoporosis.Recent studies have revealed that accumulation of senescent cells in the bone microenvironment is involved in the pathogenesis of osteoporosis;and removing these senescent cells or reducing senescence-associated secretory phenotype may prevent or slow down osteoporosis.Therefor,anti-cellular senescence therapy may become a novel strategy for treatment of osteoporosis or other aging-related diseases.This article reviews the research progress on the mechanism of cellular senescence in osteoporosis and anti-cellular senescence therapy for the prevention and treatment of osteoporosis in laboratory and pre-clinical studies.
