Objective:To explore an effective method for preparing paraffin sections of new skin tissue from wounds of SD rats and mice.Methods:21 male SD rats and 21 male SD mice were used to establish a wound model.The new skin tissue of the wound was taken at different healing times to make paraffin sections.The key steps in the process of paraffin section making were controlled,including fixation,dehydration,wax immersion,embedding,slicing,dewaxing,rehydra-tion,staining,dehydration,and encapsulation.The differences of skin tissue paraffin sections at different healing times were compared.Results:The tissue sections prepared in the experiment were uniformly stained,the cells were closely ar-ranged without cracks,the nucleus was clearly stained in dark red,and the structures of fibroblasts,hair follicles,capil-laries and muscle tissue were clearly visible.With the extension of healing time,the epidermal layer of the skin became thicker,the hair follicles became more and more differentiated,and the number of fibroblasts increased.Conclusion:This method can effectively ensure the quality of paraffin sections and determine the degree of skin healing.
