Effect of Bixie Dampness-excreting Decoction on Th1/Th2/Th22 immune function in mice model with acute eczema
Objective:To investigate the therapeutic effect of Bixie Dampness-excreting Decoction on acute eczema mice model and the related mechanism of Th cell-mediated immune function. Methods:Thirty KM mice were randomly di-vided into normal group, model group and BXSST treatment group. The mice model of acute eczema was established by 2,4-dinitrochlorobenzene(DNCB).After the model was successfully established, the mice in the treatment group were given 1.3 g/ml Bixie Dampness-excreting Decoction by gavage,0.2 ml/one mouse;and the mice in the normal group and the model group were given the same amount of normal saline by gavage. The pathological changes of ear tissues of mice were observed by HE staining.ELISA was used to detect the expression levels of interleukin-22(IL-22),interleukin-4(IL-4)and interleukin-2(IL-2)in serum of mice. Results:HE stained pathological sections showed that compared with the model group, the thickness of epidermal spinous layer in the BXSST treatment group was significantly reduced, and the in-filtration of inflammatory cells in the tissue was reduced.Compared with normal group, the expression of IL-2 in serum in model group was decreased(P<0.01),and the levels of IL-4 and IL-22 were significantly increased(P<0.01).Compared with the model group, the expression of IL-2 in the BXSST treatment group was increased(P<0.01),and the levels of IL-4 and IL-22 were decreased(P<0.05).Conclusion:Bixie Dampness-excreting Decoction can improve the symptoms of acute eczema in mice by regulating the levels of IL-2,IL-4 and IL-22 cytokines.
