Effects of different solvents on extract components of Rhamnus utilis leaves(tea)and their hypo-glycemic and lipid-lowering effects in vitro
Objective:To further explore the potential medicinal value of Rhamnus utilis leaves(tea)by selecting water and ethanol as solvents to compare the contents of the active substances of Rhamnus utilis leaves(tea)in different sol-vents,and to study the effect of different solvent extraction of the active components of Rhamnus utilis leaves(tea)on the hypoglycemic and lipid-lowering in vitro.Methods:The total content of flavonoids compounds was analyzed by ultraviolet spectrophotometer.The glucose-lowering efficacy was evaluated by the inhibition rate ofα-amylase andα-glucosidase,and the lipid-lowering efficacy was evaluated by the inhibition rate of pancreatic lipase and the binding rate of cholate.Results:The order of total flavonoids content was:Rhamnus utilis tea(ethanol)>Rhamnus utilis leaves(ethanol)>Rhamnus utilis tea(water)>Rhamnus utilis leaves(water).When the concentration was 26 mg/ml,the inhibitory rates of Rhamnus utilis tea(ethanol)onα-amylase andα-glucosidase were 48.69%and 58.46%,respectively,indicating that Rhamnus utilis tea(ethanol)had better hypoglycemic effect.The inhibition rate of pancreatic lipase and cholate binding capacity of Rhamnus utilis tea(ethanol)were 49.36%and 43.85%,respectively,indicating that Rhamnus utilis tea(ethanol)had better lipid-lowering effect.Rhamnus utilis leaves and Rhamnus utilis tea both had the effect of lowering sugar and lipid,which might be closely related to their main components,flavonoids compounds.Conclusion:When the flavonoid compounds content of Rhamnus utilis tea extracted by ethanol as solvent is high(2.81 mg/g),the effect of reducing blood sugar and lipid in vitro is significant,which provides theoretical reference for developing the medicinal value of Rhamnus utilis tea.
