不同穴组电针对大脑中动脉闭塞模型大鼠海马中Gasdermin-D的影响

目的:观察电针背俞穴(肝俞、肾俞)、阳明经穴(曲池、足三里)、督脉穴(神庭、百会)对大脑中动脉闭塞模型(MCAO)大鼠神经功能、学习记忆能力及海马组织中Gasdermin-D mRNA表达的影响,探讨其可能的作用机制.方法:将30只SD大鼠随机分为假手术组、模型组、背俞穴组、阳明经穴组、督脉穴组,每组6只.采用Longa改良线栓法制作MCAO大鼠模型,造模24 h后背俞穴组、阳明经穴组、督脉穴组大鼠每天电针20 min,连续7 d,模型组、假手术组大鼠予以同等条件抓取.通过改良神经损伤严重度评分(mNSS)判断大鼠神经功能缺损状况;Morris水迷宫实验检测大鼠学习记忆能力;实时定量荧光PCR(q-PCR)检测海马组织中Gasdermin-D的 mRNA表达.结果:在mNSS评分中,造模后第1天模型组和3组电针组大鼠均较假手术组mNSS评分升高,差异有统计学意义(P<0.05);模型组和3组电针组大鼠比较,mNSS评分差异无统计学意义.造模后第7天,阳明经穴组和督脉穴组评分较模型组和背俞穴组明显降低,且低于同组造模后第1天,差异有统计学意义(P<0.05).在Morris水迷宫定向航行实验中,与假手术相比,模型组和背俞穴组在4 d的平台探索总时间均上升,阳明经穴组在第3、5天上升,差异有统计学意义(P<0.05);与模型组相比,阳明经穴组和督脉穴组在4 d的平台探索总时间均降低,背俞穴组在第3、6天降低,差异有统计学意义(P<0.05);与背俞穴组相比,阳明经穴组和督脉穴组在4 d的平台探索总时间均降低,差异有统计学意义(P<0.05);与阳明经穴组相比,督脉穴组在4 d的平台探索总时间降低,差异有统计学意义(P<0.05).在空间探索实验中,与假手术组相比,模型组和背俞穴组穿越平台次数减少,差异有统计学意义(P<0.05);与模型组相比,阳明经穴组和督脉穴组穿越平台次数增加,差异有统计学意义(P<0.05).在q-PCR实验中,模型组和背俞穴组Gasdermin-D的mRNA相对表达量较假手术组升高,差异有统计学意义(P<0.05);阳明经穴组和督脉穴组Gasdermin-D的mRNA相对表达量较模型组和背俞穴组均降低,差异有统计学意义(P<0.05).结论:电针督脉穴组和阳明经穴组均能改善神经功能评分,减轻神经功能缺损症状,提高学习记忆能力,降低海马组织中Gasdermin-D的mRNA表达,且效果优于背俞穴组,其机制可能与下调Gasdermin-D mRNA在海马组织中的表达有关.
Effects of electropuncture at different acupoint groups on Gasdermin-D in hippocampus of middle cerebral artery occlusion model rats
Objective:To observe the effects of electropuncture at Bei Shu(Ganshu and Shenshu),Yangming Merid-ian(Quchi and Zusanli)and Du Mai(Shenting and Baihui)on nerve function,learning and memory ability and Gasdermind-D mRNA expression in hippocampus of middle cerebral artery occlusion(MCAO)model rats,and to explore its possible mechanism.Methods:Thirty SD rats were randomly divided into sham-operated group,model group,Bei Shu acupoint group,Yangming Meridian acupoint group and Du Mai acupoint group,with 6 rats in each group.The MCAO rat model was made by reforming Longa method.The rats in the Bei Shu acupoint group,Yangming Meridian acupoint group and Du Mai acupoint group were given electropuncture for 20 min every day for 7 consecutive days for on 24 h after model-ing.The rats in the model group and the sham-operated group were captured under the same conditions.The modified neu-rological severity score(mNSS)was used to evaluate the neurological impairment in rats.Morris water maze experiment was used to detect the learning and memory ability of rats.Real-time quantitative fluorescent PCR(q-PCR)was used to detect Gasdermin-D mRNA expression in hippocampus.Results:In terms of mNSS score,the mNSS score of rats in the model group and the three electropuncture groups were higher than those in the sham-operated group on the first day after model-ing,and the difference was statistically significant(P<0.05).There was no statistical significance in mNSS scores between the model group and the three electropuncture groups.On the 7th day after modeling,the scores of the Yangming Meridian acupoint group and Du Mai acupoint group were significantly lower than those of the model group and the Bei Shu acupoint group,and were lower than those of the same group on the 1st day after modeling,with statistical significance(P<0.05).In the Morris water maze directional navigation test,compared with the sham-operated group,the total time of platform explo-ration in the model group and the Bei Shu acupoint group increased on the 4th day,and the Yangming Meridian acupoint group increased on the 3rd and 5th day,with statistical significance(P<0.05).Compared with the model group,the Yang-ming Meridian acupoint group and the Du Mai acupoint group had a significant reduction in the total time of platform explo-ration on the 4th day,and the Bei Shu acupoint group had a significant reduction on the 3rd and 6th day,with statistical sig-nificance(P<0.05).Compared with the Bei Shu acupoint group,the Yangming Meridian acupoint group and the Du Mai acupoint group had a significant reduction in the total time of platform exploration on the 4th day,and the difference was statistically significant(P<0.05).Compared with Yangming Meridian acupoint group,the total exploration time of Du Mai acupoint group on the 4th day was lower,and the difference was statistically significant(P<0.05).In the space exploration experiment,compared with the sham-operated group,the number of times of crossing the platform in the model group and Bei Shu acupoint group decreased,and the difference was statistically significant(P<0.05).Compared with the model group,the number of times of crossing the platform increased in Yangming Meridian acupoint group and Du Mai acupoint group,and the difference was statistically significant(P<0.05).In q-PCR experiment,the relative expression level of Gasdermin-D mRNA in model group and Bei Shu acupoint group was higher than that in sham-operated group,and the dif-ference was statistically significant(P<0.05).The mRNA relative expression of Gasdermin-D in Yangming Meridian acu-point group and Du Mai acupoint group was lower than that in model group and Bei Shu acupoint group,and the difference was statistically significant(P<0.05).Conclusion:Electropuncture in Du Mai acupoint group and Yangming Meridian acu-point group can improve nerve function scores,alleviate symptoms of nerve function deficit,improve learning and memory ability,and reduce Gasdermin-D mRNA expression in hippocampus,and the effect is better than that in Bei Shu acupoint group.The mechanism may be related to the down-regulation of Gasdermin-D mRNA expression in hippocampus.
