Exploration of the mechanism of Sanqi in the treatment of nonalcoholic steatohepatitis based on network pharmacology
Objective:To explore the mechanism of Sanqi in the treatment of nonalcoholic steatohepatitis(NASH)by network pharmacology.Methods:TCMSP and Swiss Target Prediction databases were used to collect the effective com-ponents and their targets of Sanqi,and GeneCards database was used to screen the disease target genes of NASH.The inter-section graph of component targets and disease targets of Sanqi was obtained by Venny 2.1.0 online mapping work plat-form,and the network of Sanqi-active component-target was constructed by Cytoscape 3.7.2 software.PPI protein network was constructed through STRING 12.0 database and core target genes were screened.Gene GO function enrichment and KEGG pathway enrichment were analyzed by David database.Results:A total of 8 effective components,338 drug target genes,729 disease target genes and 75 intersection target genes were obtained through screening in Sanqi.Functional en-richment of gene GO showed that Sanqi was mainly involved in the positive regulation of transcription from RNA polymerase Ⅱ promoter,negative regulation of transcription from RNA polymerase Ⅱ promoter,cytoplasm,protein binding and other biological processes.KEGG pathway analysis showed 20 closely related pathways,including cancer pathway,PI3K-Akt signaling pathway,AGE-RAGE signaling pathway in diabetic complications.Conclusion:Sanqi plays its role in treating NASH through multi-target and multi-pathway.
