Effects of electroacupuncture with"Biao-Ben acupoints combination"at Neiguan and Zusanli on inflammatory injury in rats with chronic myocardial ischemia
Objective:To observe the effects of electroacupuncture(EA)with"Biao-Ben acupoints combination"on inflammatory injury in rats with chronic myocardial ischemia.Methods:Twenty-one Sprague-Dawley(SD)rats were ran-domly divided into a control group,a model group,and an EA group,with seven rats in each group.The model group and the EA group were injected subcutaneously with multiple doses of isoproterenol hydrochloride[5 mg/(kg·d)]for seven con-secutive days to establish a chronic myocardial ischemia rat model.After successful modeling,the rats in the EA group re-ceived EA at Neiguan and Zusanli for 21 days,once daily for 20 minutes.The control group and the model group were con-ventionally raised and handled equally without any intervention.The changes in the electrocardiogram(ECG)before and af-ter modeling and after intervention were recorded and analyzed using a PowerLab multichannel physiological recorder.The levels of inflammatory factors interleukin-17(IL-17)and interleukin-10(IL-10)in the heart tissue were detected using the enzyme-linked immunosorbent assay(ELISA)method.The expression levels of JAK1 and STAT3 mRNA in the myocardial tissues of the three groups were measured using real-time fluorescent quantitative PCR technology.Results:After model-ing,the ST segment elevation change value in the model group was significantly higher than that in the control group with a statistically significant difference(P<0.05),indicating successful modeling.After 21 days of intervention,the ST segment height change value(post-intervention-post-modelling)in the EA group was lower than that in the model group,with a sta-tistically significant difference(P<0.05).Compared with the control group,the levels of IL-17 and the relative mRNA ex-pression of JAK1 and STAT3 were increased,and the level of IL-10 was decreased in the myocardial tissues of the model group,with statistically significant differences(P<0.05).Compared with the model group,the levels of IL-17 and the rela-tive mRNA expression of JAK1 and STAT3 were decreased,and the level of IL-10 was increased in the myocardial tissues of the EA group,with statistically significant differences(P<0.05).Conclusion:Electroacupuncture with"Biao-Ben acu-points combination"may alleviate inflammatory injury and improve cardiac function by regulating the expression of IL-17 and IL-10 inflammatory factors via the JAK1/STAT3 signaling pathway.
