Study on cryptotanshinone inhibiting LPS-induced astrocytes activation in vitro
Objective:To investigate the effect of cryptotanshinone(CTS)on lipopolysaccharide(LPS)-induced acti-vation of astrocytes(AST)in vitro.Methods:Primary mice AST was isolated and cultured in vitro.CCK-8 assay was used to detect the cytotoxicity of CTS to AST.CTS did not show obvious cytotoxicity to mice AST within the concentration range of 3-30 μmol/L.In subsequent experiments,the AST were randomly divided into a control group,a model group,and three con-centrations CTS groups.The cells in the CTS administration group were pretreated with 3 μmol/L,10 μmol/L and 30 μmol/L CTS for 2 hours,and then stimulated with 100 ng/ml LPS for 24 hours.The cells in the model group were also stimulated with 100 ng/ml LPS for 24 hours,while the cells in the control group were conventionally cultured.Real-time PCR was used to detect the mRNA expression levels of cytokines interleukin-6(IL-6),tumor necrosis factor-alpha(TNF-α),interleu-kin-1 beta(IL-1β),monocyte chemoattractant protein-1(MCP-1),and inducible nitric oxide synthase(iNOS),as well as the mRNA expression levels of A1 astrocyte marker genes H2-T23,H2-D1,and Serping1.Results:Real-time PCR re-sults showed that CTS significantly reduced the mRNA expression levels of IL-6,TNF-α,IL-1β,MCP-1,iNOS,H2-T23,H2-D1,and the difference was statistically significant(P<0.05).Conclusion:CTS can effectively inhibit LPS-induced AST activation in vitro,reduce cytokine release,and inhibit the conversion to A1 type AST.
