Study on efficacy and mechanism of Lianzhi Powder on wound repair after hemorrhoids surgery
Objective:To explore the efficacy and mechanism of Lianzhi Powder(LZP)on wounds repair after hemorrhoids surgery.Methods:Twenty-four New Zealand rabbits were randomly divided into blank control group(Con group),Model group,LZP group and Mayinglong group(MHO group).Except for the Con group,wounds were inflicted on the anal pad and mixed bacterial solution was injected to establish the model of rabbit perianal ulcer.Two days after modeling,100 mg LZP powder and 100 mg Mayinglong Musk hemorrhoid ointment were applied to the wound for 5 consecutive days in LZP group and MHO group,respectively.During this period,the mass and perianal thickness of rabbits were measured,and blood samples were collected to detect number of white blood cells(WBC)and neutrophils(Neut)in the blood.After the intervention,hematoxylin-eosin(HE)staining was used to observe the histopathology at the anorectal site of rabbits,and the serum levels of interleukin-6(IL-6),interleukin-1β(IL-1β)and tumor necrosis factor-α(TNF-α)were detected by enzyme-linked immunosorbent assay(ELISA).The mRNA expression levels of TNF-α,IL-1β,IL-6,transforming growth factor-β1(TGF-β1),vascular endothelial growth(VEGF)and epidermal growth factor(EGF)were detected by RT-qPCR.The protein expression levels of VEGF,matrix metalloproteinase(MMP-9)and TGF-β1 were detected by Western blot and immunohistochemistry.Results:①Compared with the Model group,LZP could alleviate the decline of body mass(P<0.01),reduce the degree of perianal swelling(P<0.05),significantly reduce number of WBC and Neut in the blood(P<0.01),and significantly reduce the expression levels of inflammatory factors IL-6,IL-1 β and TNF-α in serum(P<0.05).②RT-qPCR results showed that compared with the Model group.mRNA expression levels of pro-inflammatory cytokines TNF-α,IL-1β and IL-6 in anorectal tissue of rabbits were significantly decreased after LZP intervention(P<0.05).The mRNA expression levels of growth factor VEGF,MMP-9 and TGF-β1 were significantly increased(P<0.05,P<0.01).③Western blot and immunohistochemical results showed that LZP intervention significantly increased the protein expression levels of growth factors VEGF,MMP-9 and TGF-β1(P<0.05,P<0.01).Conclusion:LZP can effectively promote angiogenesis and scar formation,eliminate edema and accelerate the repair speed of infectious wounds after hemorrhoidal surgery by increasing the expression level of growth factors,inhibiting inflammatory factors expression.
