Effects of Salvia Chinensis Benth Polysaccharides on Epithelial Interstitial Transformation of Colon Cancer SW620 Cells
Objective To investigate the effects of Salvia chinensis Benth polysaccharides on epithelial mesenchymal transition(EMT)in colon cancer SW620 cells,and to explore the mechanism of Salvia chinensis Benth polysaccharides in inhibiting invasion and metastasis of colon cancer.Methods Different concentrations of Salvia chinensis Benth polysaccharides were used to intervene SW620 cells for 12 and 24 hours,respectively.The proliferation of each group of cells was detected using the CCK-8 method,and the drug concentration and time for subsequent experiments were selected;Cell scratch and Transwell migration experiments were used to detect the migration ability of each group of cells;Transwell invasion experiment was used to detect the invasiveness of cells in each group;Western blot and qRT-PCR assays were used to detect the expression of EMT related proteins E-cadherin,Vimentin and their mRNA,as well as transcription factors ZEB1,ZEB2,Twist1 and their mRNA.Results Compared with the control group,Salvia chinensis Benth polysaccharides can inhibit the proliferation of SW620 cells,and the higher the concentration and the longer the intervention time,the stronger the inhibitory effect in a concentration and time dependent manner.Therefore,in subsequent experiments,0.25 mg·mL-1、1 mg·mL-1、2 mg·mL-1 and 3 mg·mL-1 were selected as intervention concentrations and 24 h as intervention time.Compared with the control group,Salvia chinensis Benth polysaccharides can inhibit the migration and invasion of SW620 cells,and the higher the concentration,the stronger the inhibitory effect,in a concentration dependent manner;Compared with the control group,Salvia chinensis Benth polysaccharides can upregulate the expression of E-cadherin and its mRNA,and the higher the concentration,the higher the expression level,in a concentration dependent manner;Simultaneously downregulated the expression of Vimentin,ZEB1,ZEB2,Twist1 and their mRNA and the higher the concentration,the lower the expression level,in a concentration dependent manner.Conclusion Salvia chinensis Benth polysaccharides can inhibit the proliferation,migration,and invasion of SW620 cells,and its mechanism may be related to upregulating the expression of E-cadherin and downregulating the expression of Vimentin,ZEB1,ZEB2,and Twist1,thereby inhibiting the EMT process.
