目的 建立柳叶白前质量标准,评价不同蜜炙方法对柳叶白前体外溶出度的影响.方法 采用高效液相色谱法.①胡萝卜苷和β-谷甾醇含量测定方法:色谱柱:Dia-monsil C18(250 mm×4.6 mm,5 μm);流动相:甲醇:0.05%磷酸水(99∶1).等度洗脱;流速1.0 mL·min-1;检测波长209 nm;进样量10 μL;柱温30℃.②葡萄糖果糖测定方法:色谱柱为Inspire NH2糖分析专用柱(250 mm×4.6 mm,5 μm);以乙腈-水(77∶23)为流动相;流速为0.8 mL·min-1;柱温为35℃;进样量10 µL;蒸发光散射检测器(ELSD):漂移管温度为100℃,载气流速为2.8 L·min-1.结果 ①结果显示胡萝卜苷和β-谷甾醇成分的测定在质量浓度范围内呈现良好的线性关系(r>0.9999),平均加样回收率为99.71%和98.83%,精密度、稳定性、重复性RSD均小于2.0%.②葡萄糖果糖建立的方法在0.05-0.5mg·mL-1(r=0.9991)范围内,呈良好的线性关系.结论 该研究含量测定方法及体外溶出度实验方法学考察结果符合要求,为制定蜜炙柳叶白前质量标准和现代研究提供了科学依据.
Objective To evaluate the effect of different honey processing methods on the dissolution of Cynanchum salicifolia in vitro.Methods High performance liquid chromatography ① Carotene and β-Determination method of sitosterol content:chromatographic column:Dia monosil C18(250 mm×4.6 mm,5 µm).Mobile phase:methanol:0.05%phosphoric acid water=99∶1.Isocratic elution;Flow rate 1.0 mL·min-1;Detection wavelength 209 nm;Injection volume 10 μL.The column temperature is 30℃.②Determination method of glucose and fructose:the chromatographic column is Inspire NH2 sugar analysiscolumn(250 mm×4.6 mm,5 μm).The mobile phase was acetonitrile water(77∶23).The flow rate is 0.8 mL·min-1;The column temperature is 35℃;Injection volume 10 μL;Evaporative light scattering detector(ELSD):the drift tube temperature is 100℃,and the carrier gas flow rate is 2.8 L·min-1.Results ①The results showed that carotene and β-the determination of sitosterol showed a good linear relationship in the range of mass concentration(r>0.9999).The average recoveries were 99.71%and 98.83%.The RSD of precision,stability and repeatability were all less than 2.0%.②The established method of glucose fructose showed a good linear relationship in the range of 0.05-0.50 mg·mL-1(r=0.9991).Conclusion The content determination method and in vitro dissolution test methodology of the study meet the requirements,which provides a scientific basis for the establishment of quality standards and modern research of honey roasted Paeonia salicifolia.
