A new record species of rust fungi on Salix matsudana in Northeast China
To identify the pathogen of rust disease on Salix matsudana, the morphological characteristics of urediospores were observed using both light microscope and scanning electron microscope. The genomic DNA of urediospores were extracted,PCR amplification was performed on the ITS and LSU of ribosomes to further construct a phylogenetic tree. The pathogen was found to cluster with Melampsora ferrinii on the phylogenetic tree.Combined with morphological identification, the pathogen was determined to be M.ferrinii Toome&Aime.M.ferrinii Toome&Aime was a new record species in the Northeast region of China, and S.matsudana was reported as a new host plant for this fungus internationally for the first time.
