Determination of β-Sitosterol in Peanuts by High Performance Liquid Chromatography
Objective:To establish a rapid analytical method for the determination of β-sitosterol in peanuts by ultrasonic-assisted extraction-high performance liquid chromatography(HPLC).The method was applied to determine the contents of β-sitosterol in different peanut varieties from Liaoning,Shandong and Henan province.Method:The samples were extracted with 85%ethanol(ethanol ∶ water=85 ∶ 15,V ∶ V)by ultrasonic,after centrifugation and removing pigments and impurities with neutral alumina,separated by Agilient ZORBAX SB-C18 column,and then detected by diode array detector,and quantitated by external standard method.Result:β-sitosterol showed a good linear relationship with peak area in the range of 1.0~100.0 mg·L-1,and the regression equation was y=10.070x+2.348(r=0.999 9).The average recoveries were 88.5%~106.9%,with RSD of 2.31%~4.73%,and the limit of detection was 0.5 mg/100 g.The method was used to detect 30 peanut samples,the content of the content of β-sitosterol in different peanuts ranged from 31.6 mg/100 g to 77.0 mg/100 g.Conclusion:The method has the advantages of simple,convenient,good repeatability,high accuracy and sensitivity,which can meet the requirements of rapid analysis of β-sitosterol in peanuts.
