摘要

目的:采用HPLC和LC-MS相结合的方法,建立了富硒葛根粉中葛根素含量的测定方法.方法:样品经 70%乙醇提取,利用岛津Inertsil ODS-SP C18 柱(250 mm×4.6 mm,5 µm)分离,以甲醇-0.3%磷酸(20∶80)为流动相,测定富硒葛根粉中葛根素含量.结果:在0.1~1.3 µg·mL-1,葛根素的线性关系良好,平均加标回收率为97.39%,RSD为 1.08%.结论:HPLC法简便、快速,重复性好,可作为富硒葛根粉中葛根素含量的测定方法.

Abstract

Objective:To establish a method for the determination of puerarin in selenium-enriched pueraria powder by HPLC and LC-MS.Method:The samples were extracted by 70%ethanol and the content of puerarin in selene-enriched pueraria powder was determined by using Shimadzu Inertsil ODS-SPC18 column(250 mm×4.6 mm,5 µm)with methanol-0.3%phosphoric acid(20 ∶ 80)as mobile phase.Result:In the range of 0.1~1.3 µg·mL-1,the linear relationship of puerarin was good,the average recovery rate was 97.39%,RSD was 1.08%.Conclusion:HPLC is a simple,rapid and reproducible method for the determination of puerarin in selenium-enriched pueraria powder.
