一测多评法测定茶饮料中咖啡因、茶多酚总量及其5种主要组分的含量

目的:建立茶饮料中咖啡因、茶多酚总量、儿茶素、没食子酸、表儿茶素、表没食子儿茶素没食子酸酯和表没食子儿茶素的一测多评法.方法:采用HPLC法,以咖啡因为内参物,计算茶多酚总量,其余 5 种多酚物质的相对校正因子,并考察相对校正因子的稳定性,比较外标法和一测多评法对茶饮料中7 个指标的测定结果.结果:6 种被测物质的相对校正因子分别为儿茶素 0.34、没食子酸 1.41、表儿茶素 0.86、表没食子儿茶素没食子酸酯0.80、表没食子儿茶素1.53、茶多酚总量 133.91.相对校正因子在不同含量样品下的RSD为 2.95%~8.78%,均小于 10%,稳定性良好.一测多评法和外标法所得茶饮料中7 个指标的含量在统计学上无明显差异.结论:建立的一测多评法准确、可靠,可为茶饮料的质量控制提供参考.
Determination of Caffeine,Total Amount of Tea Polyphenols and Their Five Main Components in Tea Beverages by Using a One Test Multiple Evaluation Method
Objective:To establish a method for the determination of caffeine,total amount of tea polyphenols,catechin,gallic acid,epicatechin,epicatechin gallate and epicatechin in tea beverages.Method:HPLC was used to calculate the total amount of tea polyphenols and the relative correction factors of other 5 polyphenols,and to investigate the stability of the relative correction factors.The results of 7 indexes in tea beverage were compared between external standard method and one test and multiple evaluation method.Result:The relative correction factors of the 6 tested substances were catechin 0.34,gallic acid 1.41,epicatechin 0.86,epicatechin gallate 0.80,epicatechin 1.53,and total tea polyphenols 133.91,respectively.RSD of relative correction factors in different samples ranged from 2.95% to 8.78%,all of which were less than 10%,indicating good stability.There was no significant difference in the contents of 7 indexes in tea beverages obtained by one test and multiple evaluation method and external standard method.Conclusion:The established method is accurate and reliable,and can provide reference for the quality control of tea beverage.
