紫斑牡丹籽粕总黄酮纯化工艺研究

选用D101、AB-8、NKA-9 型大孔树脂进行静态吸附及解析性能试验,筛选出AB-8 型大孔树脂进行动态试验研究,获得了紫斑牡丹籽粕中总黄酮的最佳纯化工艺条件.结果显示,利用AB-8 型大孔树脂在上样液浓度为 5 mg·mL-1、pH值为 5、流速为3 BV·h-1、上样液体积为5 BV条件下进行吸附,在洗脱液为 70%乙醇、洗脱液流速为 1.5 BV·h-1、洗脱液体积为 7 BV条件下洗脱,纯化后紫斑牡丹籽粕总黄酮含量由 8.54%提高到32.36%,该工艺可行性较高,可为紫斑牡丹籽粕中总黄酮纯化提供有效理论依据.
Study on Purification Process of Total Flavonoids from Paeonia rockii Seed Meal
D101,AB-8 and NKA-9 macroporous resins were selected for static adsorption and resolution performance tests,and AB-8 macroporous resin was screened for dynamic tests to obtain the optimal purification process conditions for total flavonoids in Paeonia rockii seed meal.The results show that the AB-8 macroporous resin was used for adsorption under the conditions of sample solution concentration of 5 mg·mL-1,pH value of 5,flow rate of 3 BV·h-1,and sample solution volume of 5 BV,and when the eluent is 70%ethanol,the eluent flow rate is 1.5 BV·h-1,and the eluent volume is 7 BV,the total flavonoid content of purified Paeonia rockii seed meal increases from 8.54%to 32.36%.This process has high feasibility and can provide an effective theoretical basis for the purification of total flavonoids in Paeonia rockii seed meal.
