摘要

为了对新食品添加剂吡咯喹啉醌(Pyrroloquinoline Quinone,PQQ)发酵上清液分离纯化工艺中大孔树脂吸附洗脱步骤进行优化,提升洗脱收率,本文采用XAD-1600 大孔树脂对上柱液中PQQ进行吸附和洗脱,对洗脱剂种类进行筛选,对上柱液的pH值、洗脱剂中氯化钠的浓度、洗脱温度和洗脱流速等工艺参数进行优化,计算洗脱收率.结果表明,在PQQ上柱液pH值为 3.5,洗脱剂选择磷酸盐缓冲液,磷酸盐缓冲液中氯化钠含量为1.4%,洗脱流速为2 BV·h-1,洗脱温度为20℃的条件下,PQQ洗脱收率最高(96.7%).

Abstract

In order to optimize the adsorption and elution steps of macroporous resin in the separation and purification process of the new food additive pyrroloquinoline quinone(PQQ)fermentation supernant and improve the elution yield,XAD-1600 macroporous resin was used to adsorb and eluate PQQ in the upper column liquid.The elution yield was calculated by optimizing the pH value of the upper column liquid,the concentration of sodium chloride in the eluent,the elution temperature and the elution flow rate.The results showed that under the conditions of pH value of PQQ upper column solution was 3.5,phosphate buffer was selected as elution agent,sodium chloride content in phosphate buffer was 1.4%,elution flow rate was 2 BV·h-1,and elution temperature was 20℃,the elution yield of PQQ was the highest(96.7%).
