Determination of 7 Tryptamines in Chocolate by UPLC-MS/MS
Purpose:An ultra-high performance liquid chromatography-triple quadrupole tandem mass spectrometry method was developed for the determination of 7 tryptamines in chocolate.Method:7 tryptamines in chocolate were extracted by rapid and convenient methanol ultrasonic extraction method.The samples were separated by Acquity HSS T3(100 mm×2.1 mm,1.8 μm)and detected by multiple reaction monitoring mode under positive ion mode of electrospray ion source.Result:The detection limits of 7 tryptamines in chocolate were 1.10~2.40 μg·kg-1,and the quantitative limits were 3.60~7.95 μg·kg-1,the linear relationship was good in the range of 10~1 000 μg·kg-1,and R2 was greater than 0.998 6,in the low,medium and high(10 μg·kg-1g、100 μg·kg-1、1000 μg·kg-1)concentrations,the matrix effect was 94.12%~112.13%,the extraction recovery was 89.68%~105.20%,and relative standard deviation within and between days were 1.22%~6.06%and 2.48%~7.26%,respectively.Conclusion:The method has high sensitivity and good accuracy,and can be used for rapid determination of 7 tryptamines in chocolate.
