Simultaneous Determination of Five Nucleotides in Infant Milk Powder by Ultra Performance Liquid Chromatography-Tandem Mass Spectrometry
Objective:To establish a method for simultaneous determination of five nucleotides in infant milk powder by ultra performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS).Method:The samples were dissolved in deionized water,extracted by ultrasound,and the pH value was adjusted to the isoelectric point by formic acid to precipitate the protein.The supernatant was purified by HLB solid phase extraction column and determined by UPLC-MS/MS.Result:The five nucleotides showed good linear relationships within the corresponding mass concentration range,and the correlation coefficients R2 were all greater than 0.999 0.The detection limit of the method was 0.15~0.45 mg·kg-1,the limit of quantification was 0.50~1.50 mg·kg-1,the average recovery was 86.0%~96.7%,and the relative standard deviation was 3.49%~7.81%.Conclusion:This method is simple to operate,has low solvent toxicity,high recovery,and good reproducibility.It can be used for routine batch determination of five nucleotides in infant milk powder.
