超高效液相色谱-串联质谱法同时测定婴儿乳粉中5种核苷酸

目的:建立超高效液相色谱-串联质谱法(Ultra Performance Liquid Chromatography-Tandem Mass Spectrometry,UPLC-MS/MS)同时测定婴儿乳粉中 5 种核苷酸的方法.方法:样品用去离子水溶解,超声提取,甲酸调节pH值至等电点使蛋白沉淀后,离心取上清液用HLB固相萃取柱净化,UPLC-MS/MS测定.结果:5种核苷酸在相应质量浓度范围内呈良好线性关系,相关系数R2 均大于0.999 0.方法检出限为0.15~0.45 mg·kg-1,定量限为0.50~1.50 mg·kg-1,平均回收率为86.0%~96.7%,相对标准偏差为3.49%~7.81%.结论:该方法操作简便、使用溶剂毒性低、回收率高、重现性好,可用于日常批量测定婴儿乳粉中5种核苷酸.
Simultaneous Determination of Five Nucleotides in Infant Milk Powder by Ultra Performance Liquid Chromatography-Tandem Mass Spectrometry
Objective:To establish a method for simultaneous determination of five nucleotides in infant milk powder by ultra performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS).Method:The samples were dissolved in deionized water,extracted by ultrasound,and the pH value was adjusted to the isoelectric point by formic acid to precipitate the protein.The supernatant was purified by HLB solid phase extraction column and determined by UPLC-MS/MS.Result:The five nucleotides showed good linear relationships within the corresponding mass concentration range,and the correlation coefficients R2 were all greater than 0.999 0.The detection limit of the method was 0.15~0.45 mg·kg-1,the limit of quantification was 0.50~1.50 mg·kg-1,the average recovery was 86.0%~96.7%,and the relative standard deviation was 3.49%~7.81%.Conclusion:This method is simple to operate,has low solvent toxicity,high recovery,and good reproducibility.It can be used for routine batch determination of five nucleotides in infant milk powder.
