Determination of Melatonin Content in Soft Candy by High Performance Liquid Chromatography
Objective:To establish a method for detecting melatonin in soft candy products by high performance liquid chromatography(HPLC).Method:The soft candy samples were frozen and crushed with liquid nitrogen,extracted with 0.5 g·L-1 potassium dihydrogen phosphate solution,fixed to volume with acetonitrile,analyzed by HPLC,and quantified by external standard method.Result:In the concentration range of 0.08~0.20 mg·mL-1,the method had good linearity,with a linear correlation coefficient of 0.999 9;the average spike recovery of the method was 97.84%~100.32%,and the precision was 0.24%~0.41%.Conclusion:This method has good specificity,simple operation,accurate results,and is suitable for the detection of soft candy products containing melatonin.
