目的:建立一种基于核磁共振氢谱(1H Nuclear Magnetic Resonance,1H-NMR)快速测定八角茴香中莽草酸含量的方法.方法:采用ZGPR核磁脉冲序列压制水峰,以对苯二甲酸二钠为内标,采集待测样品氢谱;对比内标物和莽草酸定量峰的响应峰面积,计算出样品中的莽草酸含量.结果:目标物质浓度在0.10~4.00 g·L-1 时线性方法关系良好,检出限为0.108 5 g·L-1,定量限为0.328 8 g·L-1,精密度及重复性良好.结论:本研究建立的氢核磁定量方法操作简单、快捷,方法可靠,可用于快速测定八角茴香中莽草酸的含量.
Quantitative NMR Study of Shikimic Acid in Aniseed Fennel
Objective:To establish a rapid method for the determination of shikimic acid in aniseed fennel based on 1H nuclear magnetic resonance(1H-NMR).Method:The water peak was suppressed by ZGPR nuclear magnetic pulse sequence,and the hydrogen spectrum of the sample was collected with disodium terephthalate as the internal standard.The content of shikimic acid in samples was calculated by comparing the response peak area of internal standard and shikimic acid quantitative peak.Result:The linear method in the range of 0.10~4.00 g·L-1 has good relationship,the detection limit was 0.108 5 g·L-1,the limit of quantification was 0.328 8 g·L-1,precision and repeatability were good.Conclusion:The hydrogen nuclear magnetic quantitative method established in this study is simple,fast and reliable,and can be used for the rapid determination of shikimic acid in aniseed fennel.
