云南松松花粉中总氨基酸的含量测定及超声破壁工艺优选

目的:优选云南松松花粉超声破壁工艺,并建立其总氨基酸的含量测定方法.方法:以天冬氨酸作为对照,采用分光光度法测定云南松松花粉中总氨基酸含量;通过单因素试验和正交试验考察超声破壁最佳试验条件.结果:线性方程为A=92.143C-0.060 3,r=0.999 5,天冬氨酸的质量浓度在2.0~10.0 μg·mL-1时与吸光度成良好的线性关系,平均回收率高达 99.6%,相对标准偏差为 1.85%(n=6);超声波破壁松花粉的最佳工艺为物料量 5 g、超声功率 700 W、超声时间 40 min,破壁率可达 98.0%.结论:建立的云南松松花粉中总氨基酸含量的测定方法准确性好;优选的破壁方法稳定可行.
Determination of Total Amino Acids and Optimization of Ultrasonic Breaking Technology in Pollen of Pinus yunnanensis
Objective:To optimize the method of the ultrasonic breaking and establish determination method of total amino acids inpollen of Pinus yunnanensis.Method:Aspartic acid was used as the control,the content of total amino acids in pollen of Pinus yunnanensis was determined by spectrophotometry method.Single factor test and orthogonal test were used to investigate the best experimental conditions for breaking walls.Result:The standard curve equation of A=92.143C-0.060 3,r=0.999 5,and a good linear correlation was found in range of 2 μg·mL-1 to 10 μg·mL-1 within the mass concentration of aspartic concentration.The average recovery was 99.9%,relative standard deviation was 1.85%(n=6).The best process for ultrasonically broken pine pollen was as follows:material volume 5.0 g,ultrasonic power 700 W,ultrasonic time 40 min,and the wall breaking rate could reach 98.0%.Conclusion:The established method for the determination of total amino acid content in pollen of Pinus yunnanensis is accurate.The optimal wall breaking method is stable and feasible.
