数字聚合酶链式反应(Digital Polymerase Chain Reaction,dPCR)技术是一种新兴的核酸绝对定量分析技术,无须标准品和绘制标准曲线,通过极限稀释样品和泊松概率分布原理就可以精准检测样本初始的DNA拷贝数,具有特异性强、灵敏度高和重复性好的特性,在食品安全核酸检测领域具有较好的发展潜力.本文介绍了dPCR技术的基本原理和特点,分析了dPCR技术在食品安全核酸检测领域的研究现状,并对其在食品安全中的应用前景进行展望,以期为促进dPCR在食品安全核酸检测领域中的应用提供一定参考.
Research Progress of Digital Polymerase Chain Reaction Technology in Food Safety Nucleic Acid Detection
Digital polymerase chain reaction(dPCR)is a new absolute quantitative analysis technology for nucleic acids,which can accurately detect the initial DNA copy number of samples by limiting dilution sample and Poisson probability distribution principle without requiring standard materials and drawing standard curves.It has the characteristics of strong specificity,high sensitivity and good repeatability,and has good development potential in the field of food safety nucleic acid detection.This paper introduces the basic principle and characteristics of dPCR technology,analyzes the research status of dPCR technology in the field of food safety nucleic acid detection,and prospects its application in food safety,in order to provide some references for promoting the application of dPCR in the field of food safety nucleic acid detection.
