Simultaneous Determination of Various Preservatives in Ass Hide Glue Cake by High Performance Liquid Chromatography
Objective:To develop a high-performance liquid chromatography detection method for simultaneous determination of four common preservative additives,salicylic acid,benzoic acid,sorbic acid,and dehydroacetic acid,in ass hide glue cake.Method:Food samples undergo pretreatment steps such as extraction and protein removal.The sample was separated using a Kromasil C18 chromatographic column,and 0.025 mol·L-1 sodium dihydrogen phosphate(containing 0.02 mmol·L-1 ethylenediaminetetraacetic acid disodium salt)was used as the mobile phase for gradient elution with methanol to optimize the separation efficiency.The experiment set the injection volume to 10 μL,controlled the flow rate at 1.0 mL·min-1,maintained the column temperature at 35 ℃,and used a diode array detector for detection at a wavelength of 230 nm.Result:The linear relationship of the four preservatives was good within the range of 20~500 ng·mL-1,with a detection limit of 1~3 ng·mL-1,the correlation coefficients were all above 0.999 5,and high sensitivity.The recovery rate of spiked samples is 98.8%to 101.8%,and the method is accurate.The relative standard deviation(RSD)is 0.78%to 2.36%,indicating good reproducibility.Conclusion:The method is easy to operate,has excellent separation efficiency,and is suitable for simultaneous detection of various antiseptic ingredients in ass hide glue cake.
