白肉灵芝水提物对H2O2诱导PC12细胞凋亡的保护作用

目的 对采自四川省凉山州会东县的白肉灵芝进行种属鉴定及抗氧化功效评价.方法 基于样本的ITS序列完成系统发育分析,采用H2O2处理PC12细胞建模,MTT法检测细胞存活率,验证白肉灵芝水提物对PC12细胞的保护作用.结果 样本与白肉灵芝Ganoderma leucocontextum同属于一个分支,相似度98%.样本水提物能显著提高H2O2处理后的PC12细胞的存活率并降低Caspase-3活性.结论 样本鉴定为白肉灵芝Ganoderma leucocontextum,对H2O2诱导的PC12细胞凋亡有保护作用,其机制可能与抑制细胞凋亡线粒体途径中Caspase-3的表达有关.
Protective effects of aqueous extracts of Ganoderma leucocontextum on H2O2-induced apoptosis in PC12 cells
Objective To identify the Ganoderma leucocontextum collected from Huidong county in Liangshan Yi Autonomous Prefecture,Sichuan,China and evaluate its antioxidant effect.Methods By DNA sequences of internal transcribed spacer (ITS),the samples were identified.Apoptosis of PC12 cells were induced by H2O2,methyl thiazolyl tetrazolium (MTT) assay was used to detect the cell viability of PC 12 cells protected by aqueous extracts of Ganoderma leucocontextum at different concentrations.Results The samples were identified as Ganoderma leucocontextum.Compared with the model group,aqueous extracts of Ganoderma leucocontextum could improved cell viability,decreased apoptotic cells and apoptosis rate,and decreased the expression of Caspase-3.Conclusion Aqueous extracts of Ganoderma leucocontextum can inhibit H2O2-induced apoptosis in PC12 cells,while the mechanism may be related to the inhibition of the expression of apoptosis-related protein Caspase-3 in the mitochondrial pathway.
