Rapid detection of imidacloprid residues in tobacco leaf by colloidal gold immunochromatographic assay
Objective To establish a rapid method for determination of imidacloprid residues in tobacco leaf by colloidal gold immunochromatographic assay.Methods Colloidal gold was prepared by sodium citrate reduction method,and it was marked imidacloprid monoclonal antibody.Then the method of colloidal gold immunochromatographic assay was built and optimized.Results The results showed that the visual detection limit of imidacloprid was 5 μtg/mL,and the reproducibility was high.The false positive condition was found,but the false positive rate was low.There was hardly any cross reaction with other commonly used compounds in tobacco.The results of gold immunochromatographic assay were well consistent with those of gas chromatography method.Conclusion The established gold immunochromatographic assay is rapid,efficient and reliable,and can meet the need of on-site rapid detection of imidacloprid residues in tobacco.Therefore it achieves biotechnology convenient and rapid detection in tobacco circulation and consumption field.
