Optimization of Extraction Technology and Properties of Polyphenols from Aronia melanocarpa and Study on Antioxidant Activity

黑果腺肋花楸中富含多酚类物质,多酚具有多种生物活性,提取其中的多酚可应用于食品加工.通过超声波辅助提取黑果腺肋花楸中的多酚类物质,采用单因素试验确定了三个显著因素,分别是料液比、温度、pH.由响应面试验得到了优化后的提取工艺,并且进一步研究了多酚的抗氧化能力.最佳提取工艺为料液比1∶53(g/mL)、温度34 ℃、pH 6、提取时间25 min,黑果腺肋花楸多酚得率为0.84 mg/g.黑果腺肋花楸多酚对1,1-二苯基-2-三硝基苯肼(DPPH)自由基、2,2'-联氨-双(3-乙基苯并噻唑啉-6-磺酸)二胺盐(ABTS)自由基、羟自由基的清除率分别为97.59%,89.41%和81.56%,表明黑果腺肋花楸多酚具有较好的抗氧化性.
Aronia melanocarpa is rich in polyphenols,which have various biological activities.Extracting polyphenols from it can be applied in food processing.By ultrasonic-assisted extraction of polyphenols from Aronia melanocarpa,three significant factors were determined by single factor test,which were solid-liquid ratio,temperature and pH.The optimized extraction process was obtained by response surface test,and the antioxidant capacity of polyphenols was further studied.The optimal extraction process was as follows:solid-liquid ratio 1∶53(g/mL),temperature 34 ℃,pH 6,and extraction time 25 min,and polyphenol yield was 0.84 mg/g.The scavenging rates of the polyphenols of Aronia melanocarpa on the free radicals of 1,1-diphenyl-2-trinitrophenylhydrazine(DPPH),2,2'-hydrazine bis(3-ethyl Benzothiazole line-6-sulfonic acid)diamine salt(ABTS)and hydroxyl radical were 97.59%,89.41%and 81.56%,respectively,indicating that the polyphenols of Aronia melanocarpa had good antioxidant properties.
