Fisetin is a kind of flavonoid polyphenol compound,which widely exists in fruits,vegetables and Chinese medicinal materials such as Cotinus and Sumac.In order to optimize the conditions of ultrasonic-assisted extraction,the effects of different organic solvents,acetone volume fraction,solid-liquid ratio,ultrasonic treatment time and extraction times on the yield of fisetin from Cotinus coggygria leaves were studied by single factor test.On the basis of single factor test,response surface method was used to optimize three factors which affected the extraction amount of fisetin,such as acetone volume fraction,solid-liquid ratio and extraction time.The results showed that the optimal extraction conditions were acetone volume fraction of 60%,solid-liquid ratio of 1∶10(g/mL),extraction time of 40 min,and repeated extraction twice.Under these conditions,the yield of fisetin was 1.948 2%,which was little different from the predicted value of 1.951 3%,indicating that the optimization of the extraction parameters of fisetin by response surface was reliable.The results of this study provide a certain experimental basis for the development and utilization of the medicinal value of Cotinus coggygria and the industrial production of the related preparation.
