Effects from Onion Quercetin on Immunoregulation in Immunocompromised Mice
Study the immunoregulation effect of onion quercetin on mouse lymphocytes. The mouse spleen lymphocytes are aseptically separated to prepare a lymphocyte suspension.Set up a blank group, levamisole group, onion quercetin low concentration group(50 mg/kg),onion quercetin medium concentration group(100 mg/kg),and onion quercetin high concentration group(200 mg/kg).The MTT method is used to detect the effect of onion quercetin on mouse lymphocyte proliferation, and the ELISA method is used to detect the effect of onion quercetin on mouse lymphocytes IL-2,IL-6,IL-12 and IFN-y(interferon-y).The influence of onion quercetin on the expression of IL-2,IL-6,IL-12 and IFN-y messenger ribonucleic acid(mRNA)in mouse lymphocytes is detected by q-PCR.The results show that in the concentration range of 50-200 mg/kg,onion quercetin can enhance immunoregulation function by inducing lymphocyte proliferation, promoting lymphocyte cytokines IL-2,IL-6,IL-12,IFN-y secretion and mRNA expression.
