己烯雌酚快检试纸条制备及其在畜禽肉检测中的应用

研制己烯雌酚试纸条并应用于畜禽肉检测.从碳酸钾用量、C线和T线划膜浓度的选择着手,确定试验所需要的最佳碳酸钾使用量、C线和T线划膜浓度等参数,初步建立畜禽肉中己烯雌酚胶体金快速检测方法(夹心法).通过单因素试验、正交试验确定检测工作液配方.结果表明,通过单因素试验确定检测工作液配方中BSA含量0.2%、海藻糖含量0.2%,通过正交试验确定检测工作液中氯化钠含量2%、酪蛋白含量0.25%、乙醇含量1.0%.从试纸条检测限、灵敏度、交叉反应率3个方面考察试纸条的技术参数是否符合要求.试验结果显示:试纸条最低检测限为1 μg/kg;试纸条灵敏度测试结果为100%;试纸条对己烷雌酚、双烯雌酚的交叉反应率为33.3%,对苯甲酸雌二醇、己炔雌二醇交叉反应率<0.1.试验研制的试纸条的各项参数满足检测要求,可用于实际检测.
Development of a Rapid Determination Strip for Diethylstilbestrol and Its Application in the Determination of Poultry Meat
Develop a rapid determination strip for diethylstilbestrol applicated in the determination of poultry meat.With the application amount of potassium carbonate,concentration of C-line and T-line,the best parameters needed in the experiment were determined.A rapid method for the determination of diethylstilbestrol in poultry meat by colloidal gold was established.The formula of detection working solution was determined by single factor and orthogonal test.The results showed that the content of BSA 0.2%and trehalose 0.2%in the working solution was determined by single factor test.The content of NaCl 0.2%,casein 0.25%and ethanol 1.0%in the working solution was determined by orthogonal test.The detection limit,sensitivity and cross reaction rate of the test strip were investigated.The test result showed that the detection limit of test paper strip was 1 μg/kg,the sensitivity was 100%,the cross-reaction rate of diethylstilbestrol and diethylstilbestrol was 33.3%,and the cross-reaction rate of estradiol benzoate and ethinyl estradiol was less than 0.1.It could be concluded that all of the parameters of detection requirements complied with the detection requirements,and it could be used as a screening method in practice.
