为综合评价不同产地黄芪质量,以黄芪甲苷含量、毛蕊异黄酮葡萄糖苷含量、总灰分、浸出物、铅、镉、砷、汞和铜共9个指标为变量对甘肃省定西市、陇南市及内蒙古自治区包头市黄芪样品进行化学计量学分析,评价黄芪品质及分类情况,并对储存4年后的原料指标成分进行对比.采用Origin Pro 2023软件进行聚类分析和主成分分析,聚类结果表明,原料与变量均被分为4类,且两者存在一定相关性.而主成分分析则提取了4个主成分,内蒙古包头市样本分布较为集中,甘肃省定西市、陇南市2个地域相近的样本分布较为相近,两省/自治区的样本能较好区分.采用SIMCA14.1软件进行正交偏最小二乘判别分析,甘肃和内蒙古黄芪可有效区分,并筛选出毛蕊异黄酮葡萄糖苷、铜和砷为两省样本的代表性差异成分.长期储存后,黄芪甲苷含量变化较小,而毛蕊异黄酮葡萄糖苷含量变化较大.黄芪储存及工业应用过程中需对毛蕊异黄酮葡萄糖苷含量变化情况加以关注.
The Quality of Astragalus membranaceus from Different Origins and the Effect of Storage on Its Components
In order to comprehensively evaluate the quality of Astragalus membranaceus from different origins,taking the content of astragaloside Ⅳ,calycosin-7-glucoside,total ash,extractum,Pb,Cd,As,Hg and Cu as variables,the samples of Astragalus membranaceus from Dingxi City,Longnan City and Baotou City are analyzed by chemometrics,the quality and classification of Astragalus membranaceus are evaluated,and the raw material components after 4 years storage are compared.Cluster analysis and PCA are carried out by using Origin Pro 2023 software,and the clustering results show that raw materials and variables are divided into 4 categories,and there is a certain correlation between them,while PCA extracts 4 principal components,the spatial distribution of samples in Baotou City,Inner Mongolia is more concentrated,the distribution of similar samples in Dingxi City and Longnan City of Gansu Province is also similar,and the samples of the two provinces can be well distinguished.The OPLS-DA is carried out by using SIMCA14.1 software,Gansu and Inner Mongolia samples can be distinguished effectively,and calycosin-7-glucoside,and Cu and As are selected as the representative differences between the two provinces.After long-term storage,the content of astragaloside Ⅳ changes little,while the content of calycosin-7-glucoside changes greatly.Attention should be paid to the changes of the content of calycosin-7-glucoside during the storage and industrial application of Astragalus membranaceus.
