Breeding of High Trehalose-Producing Strain and Optimization of Transformation Conditions
A high trehalose-producing strain was obtained by γ-Co60 mutagenesis of thermostable bacillus sp.producing trehalose from nature.Using HS-202006211109-117 selected in nature as the mother strain,the mutant strain bank was constructed by γ-Co60 mutagenic mutation.The trehalose high-yielding strains suitable for industrial production were selected from the mutant strain bank through the self-constructed trehalose high-yielding strain breeding model.The transformation conditions of high-yield strains were optimized.A mutation strain F-2119 with trehalose production potential was obtained through mutation and breeding model screening,and the trehalose yield of maltose conversion at 6 h was 14.30 mg/mL,which was increased to 3.83 times of the original strain,which was 3.73 mg/mL.The genetic stability of the strain was good,and the results of substrate concentration and transformation temperature showed that the strain had the potential of industrialization.The trehalose high-yield strain breeding model established in this paper had high throughput and sufficient screening pressure,and one mutant strain with high trehalose yield,genetic stability and good industrialization potential was successfully selected from tens of thousands of mutant strains in a short time.The trehalose yield was significantly higher than that of the original strain,and was higher than that reported in the current literature.
