Effect of Metal Ions on γ-Polyglutamic Acid Production Fermented by Bacillus subtilis
The high viscosity and low yield of γ-polyglutamic acid fermentation limited its application.In order to increase the production of γ-polyglutamic acid,the effect of metal ions on γ-polyglutamic acid production by Bacillus subtilis was studied.The effects of six metal ions on γ-polyglutamic acid production by Bacillus subtilis FRD215 were studied by single factor experiment and orthogonal experiment.The results of flask-shaking experiment showed that calcium,manganese and iron ions could promote the production of γ-PGA by Bacillus subtilis,but sodium,copper and zinc ions had no significant effect on the production of γ-PGA by Bacillus subtilis.The addition of 0.8 g/L CaCl2,0.02 g/L FeCl3·7H2O and 0.1 g/L MnSO4·H2O to the fermentation medium concurrently resulted in a γ-PGA yield of 53.34 g/L,and a significant 40.0%increase from the prior optimization.The results have guiding significance for large-scale production and application of Bacillus subtilis FRD215.
