Effects of Bamboo Salt on Constipated Mice and Its Mechanism
To investigate the alleviating effect and mechanism of bamboo salt in mouse model of slow transit constipation(STC)induced by loperamide hydrochloride,the constipation model was established by inadministration of loperamide hydrochloride(10 mg/kg)for 7 consecutive days.Starting from the 8th day,loperamide hydrochloride was inadministration for 1 h,followed by inadministration of low,medium,and high doses(0.5,1.0,1.5 g/kg).f bamboo salt,2.5 mg/kg of mosapride citrate,and normal saline.On the 14th day after the end of intragastric administration,a number of defecation granules in 6 hours were measured and the intestinal propulsion rate was calculated.The colon pathological changes were observed by hematoxylin-eosin staining,and the kits were used to determine serum levels of gastrin(Gas),substance-P(SP),and nitric oxide(NO).Western blot was used to determine the relative expression of c-kit proto-oncogene protein(c-kit),vasoactive intestinal peptide(VIP),protein kinase A(PKA),and aquaporin3(AQP3)in the colon.Use real-time quantitative PCR to determine the relative expression of stem cell factor(SCF)and cyclic adenosine monophosphate(cAMP)in colon tissue.Compared with the model group,the colonic muscle layer of STC mice in the bamboo salt medium-high dose group and the mosapride citrate group gradually thickened and goblet cells increased,which alleviated the mucosal damage caused by constipation.Compared with the model group,after the intervention of medium and high doses of bamboo salt and citric acid,intestinal propulsion rate and serum Gas and SP contents in STC mice were significantly increased,while NO contents were decreased,and the protein expression levels of c-kit,VIP,PKA and AQP3 in colon tissue and the relative mRNA expression of SCF and cAMP were promoted.There were significant differences(P<0.05 or P<0.01).Compared with the model group,the colonic muscle layer of STC mice in the bamboo salt medium-high dose group and the mosapride citrate group gradually thickened and goblet cells increased,which alleviated the mucosal damage caused by constipation.Compared with the model group,after the intervention of medium and high dose of bamboo salt and citric acid,intestinal propulsion rate and serum Gas and SP contents in STC mice were significantly increased,while NO contents were decreased,and the protein expression levels of c-kit,VIP,PKA and AQP3 in colon tissue and the relative mRNA expression of SCF and cAMP were promoted.There were significant differences(P<0.05 or P<0.01).Bamboo salt has a laxative effect on STC mice induced by loperamide hydrochloride,which may achieve the effect of relieving constipation by regulating SCF/c-kit and VIP/cAMP/PKA/AQP3 signaling pathways,and this study provides a theoretical basis and scientific basis for the development of related functional foods.
