Analysis of Monosaccharide Constituent in Flos Hibiscus Polysaccharide by Precolumn Derivatization
Analyze the monosaccharide constituent of Hibiscus polysaccharide by pre-column derivatization-HPLC,and build a approach for the assay of xylose,galactose,arabinose,glucose and glucuronic acid.The condition of liquid chromatography,polysaccharide hydrolysis and derivatization was investigated and optimized.By comparing the peak areas of monosaccharide derivatives,the assay approach of monosaccharide content in polysaccharide hydrolysate of Hibiscus was determined.The results showed that HPLC conditions was that C18 column,column temperature of 30 ℃,and mobile phase of 0.1 mol/L ammonium acetate solution-acetonitrile(80+20,V/V).Hydrolysis condition was that hydrochloric acid solution of 1 mol/L,hydrolysis temperature of 110 ℃ and hydrolysis time of 60 min.Derivatization condition was that derivatization temperature of 70 ℃ and derivatization time of 90 min.The approach was simple and reproducible,and could be used to determine the monosaccharide content in polysaccharide of Hibiscus.
