Optimization of Extraction Process and Antioxidant Activity Analysis of Flavonoids from Eurycoma longifolia Jack by Response Surface Methodology
The effect of ultrasonic-assisted extraction on the content of total flavonoids in Eurycoma longifolia Jack was studied,including particle size,ratio of solid to liquid,extraction time,extraction times,extraction temperature and ethanol concentration.On this basis,the Box-Behnken method was used to optimize the extraction process of total flavonoids from Eurycoma longifolia Jack,and the antioxidant activity in vitro was evaluated by determining the free radical scavenging rate of DPPH and ABTS.The results showed that the optimal extraction conditions of the total flavonoids from Eurycoma longifolia Jack were as follows:raw material size 0.425-0.770 mm,extraction times 2 times,extraction temperature 50 ℃,extraction time 26 min,ethanol concentration 41%,ratio of material to liquid 1∶11(g/g),and the content of flavonoids was 117.33 mg/g.The IC50 of flavonoids from Eurycoma longifolia Jack on DPPH and ABTS free radical scavenging rates were 0.19 mg/mL and 0.18 mg/mL,respectively,indicating strong in vitro antioxidant capacity.
