利用物理化学手段建立常见致病性沙门菌不同血清型的基质辅助激光解吸/电离飞行时间质谱(matrix assisted laser desorption/ionization time of flight mass spectrometry,MALDI-TOF MS)指纹图谱,为食品检验、人畜禽感染诊断、疾病预防的快速鉴定病原菌的应用提供参考.对经过血清分型鉴定的8株标准菌株,利用飞行时间质谱技术,通过谱图的采集,建立各个不同血清型的质谱指纹图谱.对易受沙门氏菌微生物污染的食品禽肉、禽蛋和畜肉进行检测分析,并对标准方法及新建方法的检测鉴定分型结果进行比较.结果表明,随机选取的样品筛选得到7个不同分型沙门菌,对于建立谱库的沙门菌基本能够快速鉴定到相应血清分型,且鉴定结果与血清分型结果一致.常见沙门菌的指纹图谱的建立,可为易受沙门菌污染的食品检验工作提供快速、简便、高通量、准确的沙门菌的血清分型鉴定方法.
Establishment and Application of MALDI-TOF MS Fingerprint of Salmonella
The matrix-assisted laser desorption/ionization time-of-flight mass spectrometry(MALDI-TOF MS)fingerprints of different serotypes of common pathogenic Salmonella were established by physical and chemical methods,it can be used as a reference for food inspection,diagnosis of human,livestock and poultry infection and rapid identification of pathogenic bacteria for disease prevention.The time-of-flight mass spectrometry was used to establish the fingerprints of different serotypes of 8 standard strains.The poultry meat,poultry eggs and meat which were easily contaminated by Salmonella were detected and analyzed,and the accuracy of the standard method and the new method were compared.Results showed that randomly selected samples were screened to get 7 different types of Salmonella,for the establishment of the library of Salmonella can quickly identify the corresponding serotypes,and identification results are consistent with serotyping results.The establishment of common salmonella fingerprints can provide a rapid,simple,high-throughput and accurate method for serotype identification of Salmonella in food inspection.
