The Effects of Tyrosinase Activity of Mouse Melanoma Cell B16 on Saxifraga Extracts
The extracts of Saxifraga with different polar solvents were prepared,and their effects on the proliferation and tyrosinase activity of mouse B16 melanoma cells were studied in this paper.The samples were used to intervene mouse melanoma cell B1 6.The morphology and quantity of mouse B16 melanoma cells were observed by inverted microscope.The cell viability was detected by thiazolyl blue(MTT)method,and tyrosinase activity in B16 melanoma cells were determined by dopa oxidation method.The results showed that different solvent extracts of Saxifraga could inhibit the proliferation of B16 cells in a concentration dependent manner.Compared with the control group,the corresponding concentrations of Saxifraga samples 1-4 were 74.52,103.48,109.60 and 65.54 μg/mL,respectively,when half inhibition(IC50)of cell proliferation activity was produced.And sample 4 showed the strongest inhibition of cell proliferation.All Saxifraga samples showed inhibitory effect on tyrosinase activity in B16 melanoma cells,and the IC50 values of samples 1-2 and 4-5 were 163.79,107.36,362.64 and 307.50 μg/mL,respectively,and sample 2 had the best inhibition ability on tyrosinase activity in cells.Based on the above research,we believe that the different polar solvent extracts of Saxifraga can inhibit the proliferation and tyrosinase activity of mouse B16 melanoma cells.
