建立离子色谱-脉冲安培检测器测定婴幼儿配方乳粉中低聚果糖含量的方法.样品用80 ℃热水提取15 min,样液直接用果聚糖酶酶解(酶解温度40 ℃,酶解时间45 min,沸水浴灭酶5 min),CarboPac PA10色谱柱分离,以氢氧化钠-水为流动相,梯度洗脱,脉冲安培检测器进行检测.果糖在0.200 0~10.00 mg/L质量浓度范围内线性关系良好,R=0.999 9,质控样QC-IP-715测定结果准确,相对标准偏差(n=6)为0.37%,市售30批次婴幼儿配方乳粉低聚果糖的测定值与产品标示值相对标准偏差在0.4%~8.4%之间.该方法能高效、准确、可靠地检测婴幼儿配方乳粉中低聚果糖的含量.
Determination of Fructooligosaccharide in Infant Formula Milk Powder by Ion Chromatography
A method for the determination of fructooligosaccharide content in infant formula milk powder was established using ion chromatography-pulsed amperometric detector.The sample was extracted with hot water at 80 ℃ for 15 min.The sample solution was directly digested by fructanase(enzymatic hydrolysis temperature 40 ℃,enzymatic hydrolysis time 45 min,boiling water bath 5 min),separated by CarboPac PA10 column,using a mobile phasecomposed of a mixture of sodium hydroxide and water by gradient elution,detected by pulsed amperometric.Fructose calibration curves were linear in the concentration range of 0.200 0-10.00 mg/L,R=0.999 9.The quality control sample of QC-IP-715 results was accurate and relative standard deviation of 0.37%(n=6).The relative standard deviation between the measured value of fructooligosaccharides and the labeled value of 30 batches of infant formula milk powder on the market was between 0.4%and 8.4%.The method was efficient,accurate,reliable,and thus suitable for the determination of fructooligosaccharide content in infant formula milk powder.
