Evaluation of Measurement Uncertainty in the Determination of Sedumycin in Chicken samples by HPLC-MS/MS
As a common drug for the treatment of chicken coccidiosis,the determination of saidumycin has attracted little attention,and the evaluation of uncertainty in the determination of saidumycin in chicken meat has not been reported.The content of sedamycin in chicken was determined by liquid chromatography-tandem mass spectrometry(HPLC-MS/MS),and a mathematical model was established.The source of measurement uncertainty was analyzed by fishbone diagram.The measurement uncertainty was evaluated from six aspects of standard solution preparation,sample weighing,sample pretreatment process,instrument stability,sample repeatability detection and recovery rate.The uncertainty components were sorted according to the contribution rate.The results showed that the expanded uncertainty was 6.2 μg/kg,k=2,when the addition level of Saidumycin in chicken was 50.0 μg/kg.The main sources affecting the measurement uncertainty were the preparation of standard solution,repeatability test and recovery rate during the test.
