To provide an experimental basis for developing Dendrobium officinale flower(DOF),the solvent and extractant for DOF extraction based on DPPH scavenging rate were optimized. The antioxidant and anti-inflammatory activities of the extracted DOF were then determined. The results indicated that the optimized extraction method for DOF involved reflux extraction with 50%ethanol for 2 hours followed by extraction with n-butanol.High-performance liquid chromatography revealed the presence of flavonoids including taxifolin(0.25±0.08 mg/g),rutin(26.86±7.44 mg/g),myricetin(0.16±0.02 mg/g),and quercetin(0.31±0.06 mg/g)in the DOF extract. The DPPH scavenging rates for 0.5,0.4,and 1.0 μg/mL of the DOF extract against DPPH radical, hydroxyl radical, and reducing power could reach the higest lovel 94.45%,51.78%,and 90.74%respectively.Additionally,ELISA assay demonstrated that 1%DOF extract inhibited the production of inflammatory cytokines TNF-α and IL-6 in lipopolysaccharide-induced macrophages. In conclusion,a DOF extract with strong antioxidant and anti-in-flammatory activities was prepared, showing potential for use in functional foods and daily chemical products.
