Study on Antioxidant and Anti-inflammatory Activities of Dendrobium officinale Flower Extract
To provide an experimental basis for developing Dendrobium officinale flower(DOF),the solvent and extractant for DOF extraction based on DPPH scavenging rate were optimized.The antioxidant and anti-inflammatory activities of the extracted DOF were then determined.The results indicated that the optimized extraction method for DOF involved reflux extraction with 50%ethanol for 2 hours followed by extraction with n-butanol.High-performance liquid chromatography revealed the presence of flavonoids including taxifolin(0.25±0.08 mg/g),rutin(26.86±7.44 mg/g),myricetin(0.16±0.02 mg/g),and quercetin(0.31±0.06 mg/g)in the DOF extract.The DPPH scavenging rates for 0.5,0.4,and 1.0 μg/mL of the DOF extract against DPPH radical,hydroxyl radical,and reducing power could reach the higest lovel 94.45%,51.78%,and 90.74%respectively.Additionally,ELISA assay demonstrated that 1%DOF extract inhibited the production of inflammatory cytokines TNF-α and IL-6 in lipopolysaccharide-induced macrophages.In conclusion,a DOF extract with strong antioxidant and anti-in-flammatory activities was prepared,showing potential for use in functional foods and daily chemical products.
