Determination of Citric Acid in Natural Tobacco Flavor Dark Plum Powder by High-Performance Liquid Chromatography
The citric acid content in natural tobacco flavoring dark plum powder is determined by high-performance liquid chromatography(HPLC).The pretreatment conditions and chromatographic conditions are optimized.The extraction liquid is filtered by aqueous phase needle filter of 0.22 μm and separated by SunFire C18 column(250 mmx4.6 mm,5 μm).The liquid is eluted with methanol and 0.01 mol/L potassium dihydrogen phosphate(pH 2.7),detected by diode array detector,and quantified by peak area external standard method.The detection limit of the method is 4.02 mg/g,the quantitative limit is 13.40 mg/g,and the correlation coefficient is greater than 0.999 within the standard curve concentration range of 0.2-4 mg/mL.The relative standard deviation during and within the day is less than 1%(n=6).At the spiking levels of 100,200 and 500 mg/g,the spiking recovery rate is 95.00%to 103.50%.This method has simple pre-treatment,high precision,and accurate quantitative results,which can provide a basis for the determination of citric acid in dark plum powder.
