建立离子色谱-积分脉冲安培检测器(ICS-IPAD)测定银耳中单糖的分析方法。以KOH为流动相,采用Dionex Carbopac PA 20(3 × 150 mm)色谱柱,采用梯度淋洗方式同时测定银耳中的岩藻糖、半乳糖、葡萄糖和甘露糖。此次研究对银耳前处理条件进行优化,找出最佳多糖提取时间、提取温度和料液比(银耳与水的质量比)。研究结果表明,此方法4种单糖线性相关系数R为0。999 1~0。999 7,检出限为0。005~0。015 μg/mL,相对标准偏差为1。6%~2。6%,回收率为91。2%~107。3%,该方法能满足银耳中多种单糖的同时定量要求。
Determination of Monosaccharide in Tremella Tremella by Ion Hromatography-Integrated Pulse Amperometric Detector
Abstrad A method for simultaneous determination of fucose,galactose,glucose and mannose was estabhshed using the technology of ion chromatography-integrated pulse ampere-detector(ICS-IPAD).KOH was used as the mobile phase and Dionex Carbopac PA 20(3x150 mm)chromatographic column was used by gradient elution.This study optimized the pretreatment conditions of tremella tremella,and found out the best extraction time,extraction temperature and the ratio of material to liquid mass ratio of tremella tremella to water.The results showed that the linear correlation coefficient r2 of the four monosaccharides was 0.999 1-0.999 7,the detection limit was 0.005-0.015 μg/mL,the relative standard deviation was 1.6%-2.6%,and the recovery rate was 91.2%-107.3%.The method is capable of simultaneously quantifying multiple monosaccharides in Tremella fuciformis,fulfilling the requirement for comprehensive quantitative analysis.
